Is the specific directory within the Domino data directory on the server, and are you opening the file via the server? Or have you put it elsewehere on the network and are attempting to open it as a 'local' file ?

If it is within the Domino data directory, try using a Notes 6 Admin client or the browser based admin client to compact the file & see if that works.
